St. Luke's Theatre is a 174-seat Off-Broadway theatre at St. Luke's Lutheran Church at 308 West 46th Street, on Restaurant Row, just west of Eighth Avenue in Manhattan's Theater District.

It was operated by Edmund Gaynes and West End Artists Company from 2006 until 2020. Prior to 2006, Entertainment Events managed the space for the church.

St. Luke's Theatre was founded by former Pastor Dale Hansen, in the early 1990s.

The theatre and office spaces were closed for renovation during the COVID-19 pandemic in New York City
