Dylan Alcott defeated Niels Vink in the final, 7–5, 6–2 to win the wheelchair quad singles tennis title at the 2021 US Open. Alcott completed the Golden Slam with the win.

Sam Schröder was the defending champion,[1] but lost in the quarterfinals to Vink.
