Muchová debut at the WTA Tour in 2016.[1] Her first breakthrough was at the US Open in 2018, when she as qualifier made her Grand Slam debut and defeated former world No. 1 Garbiñe Muguruza in order to reach third-round.[2] The following year, she reached her first WTA singles final at the Prague Open in May 2019, but lost to Jil Teichmann in three-sets.[3] This led her into the top 100 for the first time.[4] Later same year, she reached her first Grand Slam quarterfinal at the 2019 Wimbledon, after stunning win over her compatriot and world No. 3 Karolína Plíšková. That was her first top 10 win.[5] In September 2019, she won her first WTA title at the Korea Open, after defeating Magda Linette in straight-sets.[6] In October 2019, she reached her first Premier-level semifinal at the Kremlin Cup.[7] All these results helped her qualify for the WTA Elite Trophy. She passed round-robin group, after wins over Sofia Kenin and Alison Riske,[8] but then lost to Aryna Sabalenka in the semifinal-match.[9] She then finished year as world No. 21.[10]
After having a start slow of the 2020 season, Muchová made a progress at the US Open, where she reached Round of 16, but then lost to later finalist Victoria Azarenka.[11] There she also defeated Venus Williams in the first round.[12] At the 2021 Australian Open, Muchová reached her first Grand Slam semifinal, after defeating world No. 1 Ashleigh Barty.[13] There she also recorded another top 10 win over Karolína Plíšková.[14]
